About Tirzepatide

Tirzepatide is a synthetic lipidated peptide: a linear chain of thirty-nine amino acid residues conjugated to a C20 dicarboxylic acid. Its sequence is based on that of GIP, and by receptor class it is a GIP and GLP-1 receptor agonist peptide.

The catalogue lists CAS number 2023788-19-2, the molecular formula C225H348N48O68 and a molecular weight of 4813.45 g/mol, covering the peptide chain and the diacid together. The diacid gives the molecule an amphiphilic character, a long hydrocarbon chain on an otherwise polar peptide, so it is strongly retained in reversed-phase HPLC. Identity is confirmed by electrospray mass spectrometry, deconvoluted from multiply charged ions to a neutral mass for comparison with the expected value. Each presentation is supplied as a lyophilised powder in a sealed vial.

Tirzepatide structure and analytical profile

In one-letter code the chain reads Y-Aib-EGTFTSDYSI-Aib-LDKIAQKAFVQWLIAGGPSSGAPPPS-NH2: thirty-nine residues from an N-terminal tyrosine with a free amine to a C-terminal serine amide, Ser39-NH2. The amide closes the chain without a free carboxylic acid, which leaves the diacid's terminal carboxyl and the acidic side chains as the molecule's acid groups.

Positions 2 and 13 carry α-aminoisobutyric acid (Aib), an α,α-disubstituted residue with two methyl groups on its α-carbon and no stereocentre. Each Aib is 14 Da heavier than alanine, so tandem mass spectrometry locates both from the fragment ions. The single tryptophan, at position 25, and the tyrosines give the chain usable ultraviolet absorbance near 280 nm.

The side chain of lysine 20 is acylated with a C20 fatty diacid, joined through a γ-glutamic acid unit and two OEG spacers, short ethylene-glycol-based amino acids. From the lysine outward the group runs spacer, spacer, γ-glutamic acid and then the twenty-carbon chain, which ends in a free carboxylic acid. The molecular formula C225H348N48O68 counts the chain, both spacers, the linker and the diacid as one molecule.

The hydrocarbon segment is what dominates the chromatography: tirzepatide is retained strongly in reversed-phase HPLC, and a chain without its diacid, or with the group on a different lysine, elutes elsewhere and weighs differently. That gives retention time and mass two independent checks on the attachment.

Of the chain's two lysines, Lys16 keeps a free side-chain amine and Lys20 carries the lipid group, so the position of the diacid is fixed in the sequence and confirmed by the fragment ions around residue 20.

The monoisotopic mass is 4810.52 Da and the average molecular weight 4813.45 g/mol. The [M+H]+ ion is calculated at m/z 4811.53, while electrospray shows mainly multiply charged ions, such as [M+3H]3+ near m/z 1604.51 and [M+4H]4+ near m/z 1203.64, which are deconvoluted to one neutral mass. Reversed-phase HPLC reports purity as the main peak's share of total peak area, and LC-MS confirms the mass.

The glutamines at positions 19 and 24 are the residues most able to deamidate, a shift of +0.98 Da that high-resolution mass spectrometry resolves from the main ion.

The material is a white to off-white lyophilised solid in a sealed vial. The sealed vial is stored at 2–8 °C short term, or at −20 °C and below for long-term storage, protected from light and away from repeated freeze-thaw cycles.

Y-Aib-EGTFTSDYSI-Aib-LDKIAQKAFVQWLIAGGPSSGAPPPS-NH2. Aib is written out because α-aminoisobutyric acid has no one-letter code, the -NH2 marks the C-terminal serine amide, and the lysine at position 20 carries the fatty diacid side chain.

4810.52 Da, against an average molecular weight of 4813.45 g/mol. The [M+H]+ ion is calculated at m/z 4811.53; in electrospray LC-MS the [M+3H]3+ ion near m/z 1604.51 and the [M+4H]4+ ion near m/z 1203.64 are the ones usually read.

Thirty-nine. Thirty-seven are coded amino acids and two, at positions 2 and 13, are α-aminoisobutyric acid. The chain runs from a free N-terminal tyrosine to a C-terminal serine amide.

On the side-chain amine of lysine 20. A C20 fatty diacid is joined there through a γ-glutamic acid unit and two OEG spacers. The formula C225H348N48O68 includes that whole side chain.

2023788-19-2. The same record gives the molecular formula C225H348N48O68 and a molecular weight of 4813.45 g/mol, and PubChem lists the compound as CID 166567236.

Because of the C20 diacid on lysine 20. The long hydrocarbon chain binds strongly to the non-polar stationary phase, so the peptide needs a higher organic fraction in the mobile phase to elute than a chain of the same length without the diacid.

A white to off-white lyophilised powder, one sealed vial per presentation, with the net peptide content printed on the label. The vial is kept sealed at 2–8 °C short term, or at −20 °C and below for long-term storage, away from light.
