About BPC-157

BPC-157 is a synthetic linear pentadecapeptide: fifteen amino acid residues, H-Gly-Glu-Pro-Pro-Pro-Gly-Lys-Pro-Ala-Asp-Asp-Ala-Gly-Leu-Val-OH, with a free amine at the N-terminus and a free carboxylic acid at the C-terminus. The sequence corresponds to a fragment of a larger peptide from gastric juice. Four of the fifteen residues are proline, three of them consecutive, and the chain carries three acidic side chains against a single lysine.

There is no cyclisation, lipidation or non-proteinogenic residue, so identity rests on sequence and mass: CAS number 137525-51-0, the molecular formula C62H98N16O22 and a molecular weight of 1419.53 g/mol. With no aromatic residue, the peptide absorbs mainly in the peptide-bond region around 214 nm, so reversed-phase HPLC with low-wavelength ultraviolet detection gives its profile and mass spectrometry confirms the expected mass. Each presentation is supplied as a lyophilised powder in a sealed vial.

BPC-157 structure and analytical profile

In one-letter code the chain reads GEPPPGKPADDAGLV, from Gly1 to Val15. Its fifteen residues are all coded L-amino acids apart from glycine, which has no stereocentre, and they fall into a few clear groups: four prolines (positions 3, 4, 5 and 8), three glycines (1, 6 and 13), two alanines, two aspartic acids, and one each of glutamic acid, lysine, leucine and valine.

The Pro-Pro-Pro run at positions 3 to 5 is the chain's most distinctive feature. Prolines lock the backbone angle at each of those positions, and the peptide bonds in front of them can sit in the cis or the trans form, which is why a proline-rich chain can show a slightly broadened main peak in HPLC without any change in composition.

Glycines at positions 1, 6 and 13 give the chain three flexible points, and the two alanines and the closing Leu-Val pair supply its only aliphatic side chains besides proline. The sequence has no cysteine, methionine or tryptophan, so it carries no disulfide bridge and no easily oxidised residue.

Both ends are unmodified: Gly1 keeps a free amine and Val15 a free carboxylic acid, so there is no acetyl cap and no C-terminal amide. With three acidic side chains, Glu2, Asp10 and Asp11, plus the C-terminal carboxyl, against one lysine and the N-terminal amine, the calculated net charge near pH 7 is about −2.

That acidic character shapes the analysis. In positive-mode electrospray the chain gives singly and doubly protonated ions, while the low pH of a typical reversed-phase mobile phase protonates the carboxyl groups during HPLC, so the chain runs as a neutral-to-cationic species on the column.

The monoisotopic mass is 1418.7042 Da, so the [M+H]+ ion is expected at m/z 1419.7114 and the [M+2H]2+ ion at m/z 710.3594. A deletion sequence missing one proline would sit 97.05 Da lower, and one missing a glycine 57.02 Da lower, so the mass spectrum separates the full chain from the likeliest synthesis by-products at once.

The formula C62H98N16O22 gives an average molecular weight of 1419.53 g/mol, about 0.83 Da above the monoisotopic value, because the heavier isotopes of carbon, hydrogen, nitrogen and oxygen are averaged in.

HPLC at around 214 nm gives purity as the main peak's share of total peak area, at ≥99% (HPLC) for this product. The material is a white to off-white lyophilised solid in a sealed vial. The sealed vial is stored at 2–8 °C short term, or at −20 °C and below for long-term storage, protected from light and away from repeated freeze-thaw cycles.

Yes. BPC157 and BPC 157 are other spellings of BPC-157: the same fifteen-residue peptide, GEPPPGKPADDAGLV, with CAS number 137525-51-0 and the formula C62H98N16O22.

1418.7042 Da, calculated from the formula C62H98N16O22. In positive-mode electrospray the [M+H]+ ion is expected at m/z 1419.7114 and the [M+2H]2+ ion at m/z 710.3594. The average molecular weight, 1419.53 g/mol, sits about 0.83 Da higher because it averages in the heavier isotopes.

About −2. Three acidic side chains (Glu2, Asp10 and Asp11) and the free C-terminal carboxyl outweigh the single lysine and the N-terminal amine, and the chain has no histidine or arginine. The figure comes from the standard side-chain pKa values of the residues in the sequence GEPPPGKPADDAGLV.

Four, at positions 3, 4, 5 and 8. The first three form a Pro-Pro-Pro run, which gives the chain its rigid N-terminal segment and can broaden the HPLC main peak slightly through cis and trans peptide bonds. They account for four of the fifteen residues, more than a quarter of the chain.

Because it has no tryptophan, tyrosine or phenylalanine, so it barely absorbs at 280 nm. At around 214 nm every peptide bond absorbs, which gives a strong, quantifiable signal for the main peak and its related substances.

BPC-157 is catalogued in the Peptide Fragments research area, where compounds are grouped by molecular class. The research area page lists each of them with its identifiers.
