MOTS-c is a synthetic linear peptide of sixteen amino acid residues, H-Met-Arg-Trp-Gln-Glu-Met-Gly-Tyr-Ile-Phe-Tyr-Pro-Arg-Lys-Leu-Arg-OH, and a mitochondria-derived peptide. It is built only from standard amino acids, with a free amine at the N-terminus, a free carboxylic acid at the C-terminus and no side-chain modification. Four residues are basic (three arginines and a lysine) and four aromatic (a tryptophan, two tyrosines and a phenylalanine).
The catalogue lists CAS number 1627580-64-6, the molecular formula C101H152N28O22S2 and a molecular weight of 2174.6 g/mol; the two sulfur atoms come from its two methionines. Those methionines are the likeliest sites of oxidation, each adding 16 Da, so mass spectrometry looks for them alongside the expected mass, while the tryptophan and tyrosines give good ultraviolet absorbance at 280 nm for HPLC. Each presentation is supplied as a lyophilised powder in a sealed vial.
In one-letter code the chain reads MRWQEMGYIFYPRKLR, from Met1 to Arg16. All sixteen residues are coded L-amino acids apart from glycine at position 7. Methionine sits at positions 1 and 6, the aromatic residues at 3 (Trp), 8 and 11 (Tyr) and 10 (Phe), and the basic residues at 2, 13 and 16 (Arg) and 14 (Lys); Gln4, Glu5, Ile9, Pro12 and Leu15 complete the chain.
The aromatic block in the middle of the chain, Tyr-Ile-Phe-Tyr, and the arginine-rich C-terminal end give MOTS-c a clear two-part character: a hydrophobic core flanked by charged residues. That mix of aromatic and basic side chains is what makes the peptide easy to see by ultraviolet detection and easy to ionise in positive-mode electrospray.
Its three arginines carry guanidine side chains, the most strongly basic group among the coded amino acids, which keeps them protonated across the whole pH range of a reversed-phase separation.
Both ends are unmodified: Met1 keeps a free amine and Arg16 a free carboxylic acid, with no acetyl cap and no amide. Against four basic side chains and the N-terminal amine, the chain has only one acidic side chain, Glu5, plus the C-terminal carboxyl.
The calculated net charge near pH 7 is about +3, so MOTS-c is a cationic peptide. Its two methionines are the main sites of oxidation: each can form the sulfoxide, adding 16 Da per site, which is why a mass spectrum is read for +16 and +32 Da companions to the main ion.
The C-terminal Arg16 keeps its free carboxyl, so the end of the chain carries both a positive side chain and a negative terminal charge.
The monoisotopic mass is 2173.1077 Da. The [M+H]+ ion is expected at m/z 2174.1150, though electrospray favours the multiply charged forms, [M+2H]2+ at m/z 1087.5612 and [M+3H]3+ at m/z 725.3765. Reversed-phase HPLC with detection at 280 nm, where the tryptophan and tyrosines absorb, or around 214 nm gives purity as the main peak's share of total peak area.
Tandem mass spectrometry confirms the order of the sixteen residues while the two methionines are read for oxidation, so one LC-MS run checks sequence, mass and the main related substances together.
The material is a white to off-white lyophilised solid in a sealed vial. The sealed vial is stored at 2–8 °C short term, or at −20 °C and below for long-term storage, protected from light and away from repeated freeze-thaw cycles.

Read the Learn articleYes. MOTSc and MOTS c are other spellings of MOTS-c: the same sixteen-residue peptide, MRWQEMGYIFYPRKLR, catalogued under CAS number 1627580-64-6.
2173.1077 Da, against an average molecular weight of 2174.6 g/mol. The [M+H]+ ion is expected at m/z 2174.1150, and in electrospray the [M+2H]2+ ion at m/z 1087.5612 and the [M+3H]3+ ion at m/z 725.3765 are the strongest.
MRWQEMGYIFYPRKLR, from the N-terminal methionine to the C-terminal arginine. Both termini are free, so the full form is H-Met-Arg-Trp-Gln-Glu-Met-Gly-Tyr-Ile-Phe-Tyr-Pro-Arg-Lys-Leu-Arg-OH.
About +3. Three arginines, a lysine and the N-terminal amine outweigh the single glutamic acid and the C-terminal carboxyl, which makes MOTS-c a cationic peptide.
The two methionines, at positions 1 and 6. Each can form a sulfoxide, 16 Da heavier, so the mass spectrum is checked for +16 and +32 Da companions to the main ion.
