About NAD+

NAD+ (β-nicotinamide adenine dinucleotide) is a dinucleotide coenzyme, not a peptide: it contains no amino acids. Its two nucleotides, nicotinamide mononucleotide and adenosine monophosphate, are joined through a pyrophosphate (diphosphate) linkage, and each is built on a D-ribose sugar. The positive charge in the name sits on the nitrogen of the nicotinamide ring, and NAD+ is the oxidised member of the NAD+/NADH redox pair.

The catalogue lists CAS number 53-84-9, the molecular formula C21H27N7O14P2 and a molecular weight of 663.43 g/mol. The adenine ring absorbs strongly at 260 nm, the usual wavelength for HPLC of NAD+, while NADH shows an additional band at 340 nm that distinguishes it spectroscopically. Nicotinamide and ADP-ribose are the expected breakdown products, and water content matters because the solid is hygroscopic. Each presentation is supplied as a lyophilised powder in a sealed vial.

NAD+ structure and analytical profile

NAD+ is built from two nucleotides. Nicotinamide mononucleotide pairs a nicotinamide ring with a D-ribose and a phosphate; adenosine monophosphate pairs an adenine base with a second D-ribose and a phosphate. The two phosphates join in a pyrophosphate bridge, P–O–P, which links the halves into one molecule with the formula C21H27N7O14P2.

The β in the full name gives the configuration at the ribose carbon that carries the nicotinamide ring. Because the formula contains phosphorus and no amino acid, NAD+ is a nucleotide coenzyme, analysed with methods for nucleotides rather than peptide sequencing.

Each D-ribose is a five-carbon sugar in its furanose ring form, joined to its base through a glycosidic bond at C1′ and to its phosphate at C5′.

In NAD+ the nicotinamide ring is a pyridinium ion: aromatic, with a positive charge on its nitrogen. The NADH form carries an extra hydride on carbon 4 of that ring, which turns it into a 1,4-dihydropyridine and gives the formula C21H29N7O14P2, two hydrogens more, at 665.4 g/mol.

The plus sign belongs to the ring nitrogen, not to the whole molecule. The two phosphate groups of the bridge each carry a negative charge in water, so near neutral pH NAD+ has a net charge of −1. The adenine ring gives both forms an absorbance band near 260 nm, and only NADH adds a second band near 340 nm.

The monoisotopic mass is 663.1091 Da, so the protonated ion, [M+H]+, is expected at m/z 664.1164 in positive-mode electrospray. HPLC with detection at 260 nm gives purity as the main peak's share of total peak area and separates NAD+ from nicotinamide and ADP-ribose, its expected breakdown products, and from NADH.

In negative-mode electrospray the deprotonated ion, [M−H]−, is expected at m/z 662.1018, and the main fragments correspond to the loss of nicotinamide and to ADP-ribose, the same pieces that appear as breakdown products.

NAD+ is a white to off-white hygroscopic solid, so water content is part of its specification and the vial stays closed until it is needed. The sealed vial is stored at 2–8 °C short term, or at −20 °C and below for long-term storage, protected from light and away from repeated freeze-thaw cycles.

NAD is the general abbreviation for nicotinamide adenine dinucleotide, and NAD+ names its oxidised form, with a positive charge on the nicotinamide ring. The vial holds NAD+, CAS number 53-84-9.

663.43 g/mol for the formula C21H27N7O14P2. The monoisotopic mass is 663.1091 Da, so the [M+H]+ ion is expected at m/z 664.1164 in positive-mode electrospray. In negative mode the [M−H]− ion is expected at m/z 662.1018.

53-84-9, the number CAS Common Chemistry records for β-nicotinamide adenine dinucleotide, with the formula C21H27N7O14P2. PubChem lists the compound as CID 5892.

About −1 near neutral pH. The nicotinamide ring nitrogen carries one positive charge, and the two phosphate groups of the pyrophosphate bridge carry one negative charge each.

NADH has two more hydrogen atoms, C21H29N7O14P2 at 665.4 g/mol, from the hydride on carbon 4 of the nicotinamide ring and an extra proton. That turns the aromatic pyridinium ring into a 1,4-dihydropyridine.

Its adenine ring absorbs strongly near 260 nm, which gives HPLC a clear signal for the main peak. NADH absorbs there too, but only NADH has a second band near 340 nm, so the two forms are easy to tell apart. Nicotinamide, a breakdown product, absorbs near 260 nm too, and HPLC separates it from the main peak.

The solid is hygroscopic and takes up water from air, which is why water content is part of its specification. The vial stays sealed at 2–8 °C short term, or at −20 °C and below for long-term storage, away from light.
